Description: This excellent and much needed book describes the practical aspects of mitochondrial analysis. Methods are presented in sufficient detail and the appendixes contain much useful information.
Purpose: The editors' objective is to bring together both classical and modern methods of mitochondrial analysis to enable a broad spectrum of researchers to study mitochondrial structure and function. These are very worthy objectives because fundamental biological processes involving mitochondria are currently studied by investigators with highly varied backgrounds ranging from biochemistry and physiology to molecular genetics. The editors have done an admirable job of bringing together diverse methods from some of the leading experts in the field and presenting them in a fashion that will be of great utility to biomedical researchers.
Audience: This book is written for researchers at every level who are interested in analyzing mitochondrial structure and function. Both the editors and authors of the various chapters are credible authorities.
Features: The book covers essential methods of biochemical analysis including mitochondrial subfractionation, the assay of respiratory enzymes, and the analysis of mitochondrial translation products. These are of general utility for anyone who wishes to characterize mitochondrial structure and function. In addition, the book covers more specialized methods for analyzing protein import into mitochondria, measuring permeability transition, and assaying membrane potential. Finally, basic molecular genetic manipulations of mitochondria are described for both yeast and mammalian cells. The best aspect of the book is the variety of useful topics that are covered in sufficient detail. This makes it an indispensable reference for seasoned researchers in the area as well as to those whose work is only peripherally related. The appendixes contain much useful information especially concerning mitochondrial genomes, mitochondria-associated gene products and respiratory chain inhibitors.
Assessment: This is an extremely high quality offering that is really unique — I am not aware of any recent books that cover the range of useful methodologies presented. It should serve as an invaluable reference for a wide audience of biomedical researchers.
