Description: This book addresses the potential of antisense therapies to inhibit restenosis following percutaneous transluminal coronary angioplasty (PTCA).
Purpose: PTCA has become accepted as a safe and effective alternative to medical therapy and surgery for the treatment of coronary artery disease. Its success as a method of revascularization has contributed to its explosive growth, with approximately 500,000 procedures performed each year in the U.S. Restenosis has emerged as the major limitation to its efficacy. This volume summarizes the pathobiology of restenosis and the use of antisense therapies to prevent its occurrence. These are worthy objectives because of the magnitude of the clinical problem. The editor and contributors have done a beautiful job summarizing this new area.
Audience: This book is written primarily for vascular biologists and cardiologists. It would be of little interest to surgeons. The editor is a credible authority in the field and has collected an excellent group of contributors for this volume.
Features: The book begins with a definition of the clinical problem and a description of the cellular basis of restenosis, followed by a discussion of specific strategies for antisense therapy to battle this problem. There is an excellent chapter on problems with the local delivery of anti-restenotic drugs. The book ends with a summary and a look at future directions. The book is unique; it is the first volume to address the potential of antisense therapies to inhibit restenosis. The chapters on smooth muscle cell biology are superb and provide excellent bibliographies. The final chapter on future directions provides an excellent summary. The varying antisense therapies are well described. The principal shortcomings of this book are the lack of uniformity of style between the chapters, the poor quality of the illustrations, and the changing typeset throughout the text.
Assessment: This is a very useful book that provides a unique summary of an emerging new biotechnology that has great potential for treating restenosis. It will be very useful to investigators in this field.
